If she's running OK at highway speed (no problem pulling up hills or overtaking) then I very much doubt that there's a fuel problem.. If it were fuel, then it'd be more evident at higher revs (& under load) than taking off at idle...
I think you've answered your own question when you mention wheel/tyre size.. These would be significantly larger (in rolling diameter) than the originally fitted
tyres, yes?
By going to larger
tyres, you effectively change the final drive ratio (she's higher geared), and that is always going to make it more difficult to "lift off" on an incline, especially when towing, as the turbo is "off boost" at take-off ('till ya get some revs up, that is;-))...

In my 2005 Prado I take off the 265/65x17 standard
tyres and put on 265/70x17 and I notice a difference. I was originally running 265/75x16
tyres and this made a major difference even though I was dropping down an inch in diametre the aspect ratio made the tyre larger.
