Could someone please elighten me on the exact purpose of the ball adjustment nut on a trailer?

to take up the slack with different size balls eg metric or imperial & wear.

Yeah, as Mr Freak said, it's to take up any slack, due to wear and mnaufacturing tolerances. (But it's really not meant to adapt modern 50mm items to old 2" ones, IMHO. Only 50mm SAA certified balls and couplings should be used these days.)
Just don't do it up too tight or it will cause more wear. The way to use it is: couple to the ball as usual, loosen locknut, undo the adjusting screw until you're sure it's not touching the ball, then turn the adjusting screw inwards until it just touches the ball (finger tight), hold the adjusting screw screw and tighten the locknut.
