Started to notice a slight shudder in 2000 3.0 ltr when auto changes up into overdrive. 140 000 km on clock
Not used for towing but I am concerned that this may be a
sign of things to come.

The auto sometimes suffers from this due to tailshaft balance
being out...forget the naysayers, check tailshaft balance.

cheers for the response, I will have a look at the prop shaft, although I would have expected a problem like that to occur at all speeds!

Often it is a harmonics thing. If it is a minor bealance problem, you may only notice it at a particular speed.

I suggest you think about the auto converter lockup clutch. Check to see if the engine revs change up and down in synch with the "shudder". I had a similar happening in my auto patrol.
Not a "shudder" but intermittent "slipping" of converter lockup when in overdrive. revs changed as it happened. Started round the 200,00km mark.
Had autobox rebuilt. No further problem to date.
